چکیده

We have characterized the diversity of the chicken Pi integrin family and studied the expression of individual receptors during development. The diversity of the Pi integrin family was investigated by affinity purifying the Pi integrins from a variety of adult and embryonic tissues. These purifications reveal the relative levels of expression and also the differential expression of the a subunits in those tissues. Monoclonal antibodies were generated against the prominent 'band 1' of the embryonic chicken integrins and used to characterize the expression of this a subunit in embryonic and adult tissues. This a subunit is shown to be the chicken homologue of human a5 fibronectin receptor. The chicken a5Pi integrin is the most prominent Pi integrin in the embryo and is expressed on the majority of cell types through the day 17 stage. The distribution of this receptor in the embryo closely parallels the distribution of its ligand, fibronectin. In adult tissues, expression of this receptor is greatly diminished relative to the expression of other « subunits. The cell type distribution is highly restricted: limited primarily to the vasculature and to connective tissue regions. These studies reveal a prominent role for the a5Pi integrin in embryonic cell types and a down-regulation of this receptor on many cell types during development.

Integrin heterodimer and receptor complexity in avian and mammalian cells

We report data showing that the integrin receptor complex in chickens contains several discrete heterodimers all sharing the beta 1-integrin subunit combined separately with different alpha-subunits. Using antisera to synthetic peptides based on cDNA sequences of chicken and human alpha-integrin subunits to analyze the integrin complement of avian and mammalian cells, we show that band 2 of the...

Up-Regulation of Integrinsn α2β1 and α3β1 Expression in Human Foreskin Fibroblast Cells after In-Vitro Infection with Herpes Simplex Virus Type 1

The interaction of Herpes Simplex Virus type 1 (HSV-1) with human fetal foreskin fibroblast (HFFF) cell was studied using a recent isolate of HSV-1 which was propagated in Hep-2 cells. HFFF cells were challenged with HSV-1 with a multiplicity of infection (MOI) of 1 virus/cell for 24 hours. Flow cytometric analysis demonstrated that HSV-1 challenged HFFF cells expressed increased levels of α2β1...

Regulated expression of fibronectin, laminin and related integrin receptors during the early chondrocyte differentiation.

We have investigated the expression and localization of fibronectin, laminin, and their receptors, and we used an in vitro chick chondrocyte differentiation model to define a time hierarchy for their appearance in early chondro-genesis and to determine their role in the cell condensation process. By serum fibronectin depletion/reconstitution, or GRGDSP peptide competition experiments, we show t...

Caveolin-1-dependent beta1 integrin endocytosis is a critical regulator of fibronectin turnover.

beta1 integrins are major cell surface receptors for fibronectin. Some integrins, including beta1 integrins, are known to undergo constitutive endocytosis and recycling. Integrin endocytosis/recycling has been implicated in the regulation of cell migration. However, the mechanisms by which integrin endocytosis/recycling regulates cell migration, and other biological consequences of integrin tra...

α4β1 integrin and erythropoietin mediate temporally distinct steps in erythropoiesis: integrins in red cell development

Erythropoietin (Epo) is essential for the terminal proliferation and differentiation of erythroid progenitor cells. Fibronectin is an important part of the erythroid niche, but its precise role in erythropoiesis is unknown. By culturing fetal liver erythroid progenitors, we show that fibronectin and Epo regulate erythroid proliferation in temporally distinct steps: an early Epo-dependent phase ...
